ABSTRACT

Objective@# To investigate the transcriptomic changes in primary human oral keratinocytes (pHOKs) after coculture with Prevotella melaninogenica (P.m) and to verify the changes in human oral keratinocyte (HOK) cell lines.@*Methods@# pHOK was isolated and cocultured with P.m for 0, 4 and 24 h. Total RNA was extracted, a gene library was constructed, transcriptional sequencing was performed, differentially expressed genes (DEGs) were analyzed, gene ontology (GO) pathway analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis were performed, and the validation of DEGs was performed by qRT-PCR and Western Blot in the HOK and P.m coculture cell model. @*Results @#1 788 DEGs were detected between the 4 h group and control group, including upregulated DEGs such as lymphocyte cytosolic protein 1(LCP1), keratin 7 (KRT7) and Cilia and flagella associated protein 251(CFAP251) and downregulated DEGs such as FERM, ARH/RhoGEF and Pleckstrin domain protein 1 (FARP1), WW domain containing transcription regulator 1(WWTR1) and Discoidin, CUB and LCCL domain-containing protein 2 (DCBLD2). 1 832 DEGs were detected between the 24 h group and control group, including upregulated DEGs such as LCP1, complement C1s(C1S), kynureninase (KYNU) and downregulated DEGs such as phosphoserine aminotransferase 1 (PSAT1), FARP1 and FKBP prolyl isomerase 10 (FKBP10). There were 1 090 common differentially expressed genes (cDEGs) in the 4 h and 24 h groups, including LCP1, KYNU and long intergenic nonprotein coding RNA 958 (LINC00958). The GO pathways were mainly enriched in response to lipopolysaccharide and the molecules of bacterial origin and apical part of the cell. KEGG pathway analysis revealed enrichment in the interleukin-17 (IL-17) signaling pathway, tumor necrosis factor (TNF) signaling pathway, Toll-like receptor (TLR) pathway, etc. We verified the expression of a cDEG, Myosin1B (MYO1B), and qRT-PCR and Western Blot analysis showed that MYO1B expression was significantly upregulated between the control group and the P.m cocultured group (P<0.001), and its expression followed a time-dependent and concentration-dependent manner. @*Conclusion@#P.m played an important role in the transcriptome of oral keratinocytes.

ABSTRACT

Background and Objectives@#p21, an important member of the Cip/Kip family, is involved in inhibitory effects of RUNX1b overexpression during the early stage of human hematopoiesis. @*Methods@#and Results: We established a human embryonic stem cell (hESC) line with inducible expression of p21 (p21/hESCs). Overexpression of p21 did not influence either mesoderm induction or emergence of CD34+ cells, but it significantly decreased the production of CD43+ cells and changed the expression profile of hematopoiesis-related factors, leading to the negative effects of p21 on hematopoiesis. @*Conclusions@#In RUNX1b/hESC co-cultures when RUNX1b was induced from D0, perturbation of the cell cycle caused by upregulation of p21 probably prevented the appearance of CD43+ cells, but not CD34+ cells. The mechanisms via which CD34+ cells are blocked by RUNX1b overexpression remain to be elucidated.

ABSTRACT

ABSTRACT Phytoene synthase (PSY) is the rate-limiting enzyme for carotenoid biosynthesis. To date, several studies focused on PSY genes in the context of abiotic stress responses. In this study, two phytoene synthase encoding genes, IbPSY1 and IbPSY2, were identified from a published transcriptome and bioinformatic analysis showed that they shared conserved domains with phytoene synthases from other plants. The IbPSY1 gene was cloned and carefully characterized. Digital gene expression profiling (DGE) showed that the highest transcription level of IbPSY1 was in young leaves, and the lowest level was in stems. In vivo expression levels of IbPSY1 under abiotic stress were observed to be highest in stems at day 11. Over-expression of IbPSY1 in Escherichia coli and yeast cells endowed the cells with better growth under salt and drought stress than the control cells. This study demonstrated that IbPSY1 not only played an important role in vivo, but also in E. coli and yeast to improve tolerance to salinity and drought stress. Thus, IbPSY1 may be aid in the development of transgenic plants with enhanced stress tolerance.

ABSTRACT

Objective To explore the role of Notch signaling pathway in the matrix synthesis of osteoarthritis cartilage cell and its possi -ble mechanism.Methods Selected the femoral condylar cartilage of 8 patients who were admitted into our hospital and treated with knee joint replacement as the observation group,while the normal femoral condyle cartilage of one patient with above-knee amputation was selected as the control group.The specimens were given histological examination or cell isolation culture and detection.The cultured cells were divided into 4 groups,namely the normal cartilage cells,OA cartilage cells,OA cartilage cells with γ-secretase inhibitor DAPT,OA cartilage cells with recombinant human proteins Delta4.Then immunohistochemistry and Western blot were used for detecting the expression of Notch -1,Notch-3, Jagged-1,Jagged-2 and HES5 in chondrocytes in vitro.Results The expression of Notch signaling pathway and the phenotype of cartilage cells changed in the osteoarthritis.The expression of Notch-1,Jagged-1,Jagged-2 and HES5 were activated.γ-secretase inhibitor DAPT (20 μmol/L)could inhibit the expression of Notch-1,HES5,Jagged-1,and Notch-3,while it had no obvious effect on the expreesion of Jag-ged-2.Recombinant human proteins Delta 4(100 ng/μL)could promote the expression of Notch-1,Jagged-2,and HES5,and it had no obvi-ous effect on the expreesion of Jagged-1 and Notch-3.Conclusion The expression of Notch signaling pathway of cartilage cells changed in the osteoarthritis.DLL4 can activate the Notch signaling pathway and DAPT can inhibit the Notch signaling pathway.

ABSTRACT

Objective: To observe the clinical efficacy of warm needling moxibustion plus Mulligan dynamic joint mobilization for cervical radiculopathy. Methods: A total of 60 cases with cervical radiculopathy were randomly divided into group A, group B and group C by random digital table, 20 cases in each group. The patients in three groups were treated with similar warm needling moxibustion. Group A only received warm needling moxibustion; group B was treated with warm needling moxibustion plus Mulligan dynamic joint mobilization; group C was treated with warm needling moxibustion plus cervical traction. Before the treatment and after 12 sessions of treatment, the patients in the three groups were assessed for score of pain and range of motion (ROM). Results: By 12 sessions of the treatments, the scores of pain were obviously decreased in the three groups, with statistical significances (all P<0.05), and the pain scores of group B and group C were significantly different from the score of group A (both P<0.05). ROM of the neck was increased than before the treatment in the three groups, with statistical significances (all P<0.05), and the ROM of the neck of group B was significantly different from those of group A and group C (both P<0.05). Conclusion: Warm needling moxibustion plus Mulligan dynamic joint mobilization can effectively improve the neck ROM and relieve pain in patients with cervical radiculopathy.

ABSTRACT

BACKGROUND: A large number of cytokines produce in the periodontium on the tension side during orthodontic tooth movement, participate in periodontal remodeling, and make the tooth move to its normal position, thus achieving alveolar remodeling. Heat shock protein 27 (HSP27) is reported to be involved in the periodontal remodeling. OBJECTIVE: To investigate the expression of HSP27 in the rat periodontium on the tension side during the orthodontic tooth movement.METHODS: The rat models of orthodontic tooth movement were constructed using NiTi coil spring with the force value of 50 g. The periodontal samples were collected at 1, 3, 5, 7, 10 and 14 days after exerting force to detect the expression level of HSP27 in the periodontium on the tension side.RESULTS AND CONCLUSION: Immunofluorescence staining results revealed that the expression level of HSP27 in the periodontal tissues began to increase at the 1st day after exerting force, reached the highest level at the 3rd day, and then fell to the initial level at the 5th day. To conclude, HSP27 is evoked by orthodontic force early, and it may play a role in periodontal remodeling.

BACKGROUND:Extensive research has been concentrated on the reconstruction of periodontium under orthodontic force. However, there are no reports regarding the expression of heat shock protein 70 (HSP70) in rat periodontium during orthodontic tooth movement. OBJECTIVE:To investigate the expression of HSP70 in rat periodontium during orthodontic tooth movement. METHODS:Rat orthodontic tooth movement models were established using NiTi extension spring with the force of 50 g. The orthodontic periodontium was col ected at 1, 3, 5, 7, 10 and 14 days after exerting force to detect the expression of HSP70. RESULTS AND CONCLUSION:According to immunofluorescence staining, the expression of HSP70 in the periodontal tissues was increased on day 1 after exerting force, and reached the highest level on day 5, then decreased to the initial level on day 14. These results suggest that the expression of HSP70 is evoked by orthodontic force, and it may play a role in periodontal remodeling.

ABSTRACT

Objective@#To compare the clinical effects of transperitoneal (Tp) versus extraperitoneal (Ep) robot-assisted radical prostatectomy (RARP) in the treatment of localized prostate cancer.@*METHODS@#We searched PubMed, EMBASE, Web of Science, EBSCO, Cochrane Library, Wanfang, CNKI, and CBM for the articles comparing the clinical effect Tp-RARP with that of Ep-RARP in the treatment of localized prostate cancer published from January 2000 to November 2016. All the articles must meet the inclusion criteria, that is, dealing with at least one of the following aspects: operation time, intraoperative blood loss, postoperative catheterization time, length of bed confinement, perioperative complications, positive surgical margins, bowel-related complications, postoperative anastomotic leakage, and postoperative urinary continence. We subjected the data obtained to statistical analysis using the RevMan5.3 software.@*RESULTS@#Two randomized controlled trials and six case-control studies were included in this meta-analysis, involving 451 cases of Tp-RARP and 676 cases of Ep-RARP. Compared with Tp-RARP, Ep-RARP showed significantly shorter operation time (WMD = 21.39, 95% CI: 7.54-35.24, P = 0.002), shorter length of bed confinement (WMD = 0.85, 95% CI: 0.61-1.09, P <0.001), and lower rate of bowel-related complications (RR = 9.74, 95% CI: 3.26-29.07, P <0.001). However, no statistically significant differences were found between the two strategies in intraoperative blood loss (WMD = -8.12, 95% CI: -27.86-11.63, P = 0.42), postoperative catheterization time (WMD = 0.17, 95% CI: -0.55-0.21, P = 0.38), or the rates of perioperative complications (RR = 1.34, 95% CI: -0.97-1.87, P = 0.08), positive surgical margins (RR = 1.24, 95% CI: 0.95-1.61, P = 0.12), anastomotic leakage (RR = 0.98, 95% CI: 0.46-2.10, P = 0.95), urinary continence at 3 months (RR = 0.96, 95% CI: 0.91-1.00, P = 0.05) and urinary continence at 6 months (RR = 1.00, 95% CI: 0.97-1.02, P = 0.82).@*CONCLUSIONS@#Ep-RARP has the advantages of shorter operation time, shorter length of bed confinement and lower rate of bowel-related complications over Tp-RARP, and therefore may be a better option for the treatment of localized prostate cancer. However, more multi-centered randomized controlled clinical trials are needed for further evaluation of these two approaches.

ABSTRACT

BACKGROUND:Cigarette smoking can seriously damage the periodontal tissues and root, and the nicotine in tobacco accelerates the progression of periodontal diseases. OBJECTIVE: To investigate the effect of different doses of nicotine on the expression of cyclooxygenase-2 mRNA in the periodontal tissue during orthodontic tooth movement. METHODS: Totaly 110 Sprague-Dawley rats were randomly divided into blank control group (n=10), normal saline group (n=25), 0.5 mg/kg nicotine group (n=25), 0.75 mg/kg nicotine group (n=25), 1 mg/kg nicotine group (n=25). Rats in the normal saline groups were injected intraperitonealy with 0.1 mL normal saline, and those in the three nicotine groups were respectively injected with 0.5, 0.75, 1 mg/kg nicotine tartrate solution. Except the blank control group, the unilateral maxilary first molars of rats in the other four groups were exposed to 50 g force. At 1, 3, 5, 7, 14 days under force, the rats were sacrificed to take the maxilary tissues. Hematoxylin-eosin staining was used to observe the changes of periodontal tissues, immunohistochemical staining was employed to count positive cels, andin situ hybridization staining was adopted to detect the mean absorbance value of cyclooxygenase-2 in the periodontal tissues. RESULTTS AND CONCLUSION:The number of odontoclasts and the expression of cyclooxygenase-2 in the nicotine groups were higher than those in the non-nicotine groups. With the increasing dose of nicotine, the number of odontoclasts gradualy increased, and the difference was statisticaly significant (P < 0.05). At 7 days under force, the number of odontoclasts reached the peak. With the increasing dose of nicotine, the positive expression intensity of cyclooxygenase-2 was also increased, and the difference was statisticaly significant (P < 0.05). The expression of cyclooxygenase-2 reached peak at 5 days under force. These findings indicate that with the increasing dose of nicotine, the number of odontoclasts and the expression intensity of cyclooxygenase-2 are both increased at the same time point under force. During the orthodontic tooth movement, the intake of nicotine can damage the periodontal tissue, and the dose of nicotine can directly influence the severity of damage to the periodontal tissue.

ABSTRACT

Objective To investigate the value of ultrasound guided autobiopsy technology in interventional diagnosis of pulmo-nary peripheral disease .Methods 117 cases of pulmonary peripheral disease proved by X-ray or CT ,confirmed by ultrasound ,but could not be identified the nature by the examination above ,were biopsied under the guidance of ultrasound .Results All the 117 ca-ses were biopsed successfully .115 cases of them had definite pathological results ,the pathological positivity rate was 98 .3% ,in which contained 20 cases of chronic inflammation ,14 cases of tuberculosis ,30 cases of squamous cell carcinoma ,45 cases of adeno-carcinoma ,6 cases small cell undifferentiated carcinoma .No complication ,such as pneumatothorax ,hemoptysis ,bleeding occurred during the operation .Conclusion The ultrasound guided autobiopsy technology in interventional diagnosis of pulmonary peripheral disease is a safe and effective technique ,the tissues obtained from ultrasound guided autobiopsy were adequate to pathological slide which can provide the basis of the clinical diagnosis .

ABSTRACT

BACKGROUND:As an important cytokine, interleukin-6 regulates immune responses in inflammation sites and has an autocrine/paracrine activity that stimulates osteoclast formation and bone resorption, which is related to bone remodeling during orthodontic tooth movement. OBJECTIVE:To investigate the effects of orthodontic force on the expression of interleukin-6 mRNA in the periodontal tissue of rats. METHODS:In situ hybridization was performed to measure the expression of interleukin-6 mRNA at 1, 3, 5, 7, 10 and 14 days after the application of orthodontic force on the maxil ary first molars of rats. RESULTS AND CONCLUSION:The expression of interleukin-6 mRNA was observed at a low level in the normal periodontal tissue of rats. After the application of force, the induction of interleukin-6 mRNA was observed to reach a maximum on day 3 and to decline thereafter. The expression of interleukin-6 mRNA can be evoked by orthodontic force but with a certain self-limiting. As a multifunctional cytokine, interleukin-6 plays a very important role in periodontal remodeling during orthodontic tooth movement.

ABSTRACT

This study was aimed to compare different preparation methods of Wei-Chang Fu-Yuan (WCFY) De-coction, which is a self-made traditional Chinese medicine (TCM) decoction of our hospital, through the investiga-tion of difference in recovery time of gastrointestinal function after gynecological abdominal surgery. A total of 120 cases after gynecological abdominal surgery in our hospital between 2011 and 2012 were randomly divided into the treatment group and the control group, with 60 cases in each group. The hospital self-made WCFY Decoction was given to patients 8 hours after surgery in the treatment group and control group. Decoctions given to two groups were prepared with different methods. Decoction given to the treatment group was by the decoction ma-chine of multiple decoctions preparation. Decoction given to the control group was prepared as the traditional sin-gle dose decoction. The first anus flatus time was observed in both groups. The results showed that the flatus time of treatment group was 16 h and that of the control group was 13 h. The flatus time of patients from two groups was in skewed distribution, so the distance between the median and quartile was used to describe. The non-para-metric tests (Kolmogorov-Smirnov method) were used in the comparison of flatus time of two groups. There was statistical significance between two groups (P < 0.05). It was concluded that different preparation methods affect the clinical efficacy of WCFY Decoction. Compared with the traditional single dose decoction, the fist flatus time was postponed 3 hours by the decoction machine of multiple decoctions preparation. Therefore, with the widely using of decoction machine of multiple decoctions preparation today, the function of decoction machine should be improved and the program should be perfected in order to increase the clinical efficacy of this preparation method.

ABSTRACT

Objective To share the experience of heart thransplantation. Methods 3 recipients with terminal myocardiosis were reviewed. The transplantation was performed with inferior and superior vena anastomofic technique. During perioperative period, we selected and maintained the recipients, protected donor-isolated heart, supported circulation,decreased immune reaction and controlled infections. Results All the 3 patients survived. Heart function improved from NYHA class 1V before heart transplant to NYHA class Ⅰ, Ⅱ. The follow-up time was 19 months ,28 months and 49 months respectively. Rejection occurred in two cases due to non-compliance to medication. Conclusions Suitable recipient, proper donor heart procurement and preservation, suitable maintenance of circulation, proper managements of anti-immunitive reaction, prevention of infections are critical for successfal heart transplantation. Medicine-take required may avoid or reduce rejection.

BACKGROUND:It is very important to appreciate the Sagittal discrepancy during the diagnosis and therapy in orthodontics patients.At present,ANB angle,AXB angle,Beta angle and APDI value were often used to evaluate the mandible and maxilla relationships in orthodontic clinic.Correlation and variance degree among these parameters were analyzed to judge the reliability.OBJECTIVE:To compare the reliability of ANB angle,AXB angle,Beta angle and APDI value in assessing sagittal discrepancy between mandible and maxilla in transitional dentition cases with Angle's class Ⅲ malocclusion.METHODS:Totally 56 patients of Angle's class Ⅲ malocclusion in transitional dentition at the Department of Orthodontics,First Affiliated Hospital of Xinjiang Medical University,between January 2004 and June 2009,were selected.ANB angle,AXB angle,Beta angle and APDI value of Angle's class Ⅲ malocclusion in transitional dentition were measured on the cephalometric radiographs.The radiographs were scanned by Microtek Phantom 9800 XL scanner at proportion of 1:1 with 300 dpi pixels,and saved as JPG format.The JPG pictures were fixed-point using Photoshop 8.0 to measure ANB angle,AXB angle,Beta angle and APDI value,the mean value was obtained after 3 times measurement.The correlation and variance degree among these parameters were analyzed.RESULTS AND CONCLUSION:Significant correlation was found among ANB angle,AXB angle,Beta angle and APDI value (P<0.05).The coefficients of variation for the 4 indexes were 1.35,0.75,0.07 and 0.04,respectively.The results demonstrated that,it is accurate to use ANB angle,AXB angle,Beta angle and APDI value as cephalometric norm to evaluate sagittal discrepancy of maxmandibul in transitional dentition cases with Angle's class Ⅲ malocclusion.However,the APDI value is more reliable as cephalometric norm.

ABSTRACT

Objective The facial soft tissue features were studied in Urumqi adolescent with skeletal Ⅱ malocclusion by using a polar coordinate system. Methods Sixty samples of Urumqi adolescent with skeletal Ⅱ malocclusion were selected, each one underwent cephalometrics analysis and 32 cephalometric parameters from the samples were measured. The mean and standard deviations of each measurement were compared by statistical analysis (t-test). Results Polar distance in male was larger than female, showing significant difference between male and female. Po-Ns/Po-Li, Po-Ns/Po-Bs, Po-Ns/PoPgs, Po-Ns/Po-Gns, and Po-Ns/Po-Mes showed significant difference between male and female. Polar angles in female were larger than male, with a significant difference between male and female.Conclusion Statisticalstudies on features of facial soft tissue show significant differences between male and female. The polar coordinate analysis system is a good index to study the growth magnititude and direction of facial soft tissue during growth and development or before treatment and after treatment.

BACKGROUND:Beta angle,as a new index for the assessment of sagittal jaw relationship,has been widely used by clinical doctors.However,hand-made measurement but not academic software is still adopted in some hospital.OBJECTIVE:To study the beta angle of different skeletal patterns in Urumqi city for the clinical use of the method to assess the sagittal jaw relationship,and verify the possibility of Photoshop software to measurement of beta angle.DESIGN,TIME AND SETTING:An observational experiment and measurement design was performed at Department of Orthodontics,the First Affiliated Hospital of Xinjiang Medical University from July 2006 to May 2009.PARTICIPANTS:A total of 262 malocclusion cases including 94 males and 168 females were lived in Urumqi City.They were divided into three groups based on type of facies ossea,including type Ⅰ group (n=119),type Ⅱ group (n=74),and type Ⅲ group (n=69).METHODS:Lateral position of skull was photographed using X-ray,and beta angle was measured using Photoshop 8.0 software.Sex and type of facies ossea were compared using one-way analysis of variance and t test.MAIN OUTCOME MEASURE:Beta angle of male and female;beta angle of the types Ⅰ,Ⅱ,and Ⅲ.RESULT:There was no significant difference in beta angle between males and females (P>0.05).Beta angle of type Ⅰ was (34.15±3.88)°,that of type Ⅱ was (26.42±2.65)°,and that of type Ⅲ was 42.41°±1.88°.There was significant difference in beta angle among three types of facies ossea (P<0.01).CONCLUSION:Beta angle is an index for the assessment of sagittal jaw relationship.The reference range of the beta angle is 30°-38°.Photoshop 8.0 software is a novel tool to measure cephalometry.
